Technical Notes: StemXVivo Media for Stem Cell Culture

TECHNICAL NOTES: STEMXVIVO MEDIA FOR STEM CELL CULTURE

Researchers employing stem or progenitor cells in their studies currently use a wide variety of media products to support their cultures. Unfortunately, these media are generally developed or validated for cells other than stem cells, thus making them less desirable than cell type-specific media.

To address this need, R&D Systems offers two new specialty media products for stem/progenitor cell culture. All the components included within these defined, basal, serum-free formulations have been selected, optimized, and validated for primary stem cell culture. StemXVivo Serum-free HSC Base Media is formulated for the maintenance and expansion of purified human CD34+ hematopoietic stem cells. StemXVivo Serum-free NSC Base Media is formulated for expansion and differentiation of human and rodent neural stem and progenitor cells.

For monolayer culture, cells were expanded using R&D Systems Fibronectin-coated plates (Catalog # 1030-FN) and StemXVivo Serum-Free NSC Base Media (Catalog # CCM002) supplemented with recombinant human FGF-basic (Catalog # 233-FB; 20 ng/mL ). For neurosphere culture, cells were expanded using the StemXVivo Serum-Free NSC Base Media supplemented with recombinant human FGF-basic and EGF (Catalog # 236-EG; 20 ng/mL).

Cells were cultured in R&D Systems StemXVivo Serum-Free HSC Base Media (Catalog # CCM001) supplemented with 100 ng/mL each R&D Systems recombinant human Flt-3 Ligand (Catalog # 308-FKN), SCF (Catalog # 255-SC), and Tpo (Catalog # 288-TP). On days 10 and 14, cells were harvested and assayed for TNC and CD34+ cell expansion. Viable TNCs were counted using a hemocytometer and the CD34 expression was analyzed by flow cytometry.
