Cell-Based ELISAs
Cell-Based ELISAs

R&D Systems Cell-Based ELISAs are the first fluorescence assays to permit the simultaneous screening of both phosphorylated and total proteins in the same microplate well. These kits contain the components required for running an ELISA including two different fluorogenic substrates to measure phosphorylated and total proteins. Normalizing the fluorescence signal derived from the phospho-protein to that of the total protein in the same well makes it easy to account for potential well-to-well variability such as differences in cell number. These simple and efficient assays are carried out using whole cells without the need to prepare cell lysates. Consequently, they are amenable to high-throughput investigation of kinase signaling and inhibition.

Assay Principle

*Kit Components

  • 96-Well Cell Culture Plate
  • Phospho-specific Antibody
  • Total Antibody
  • HRP-Conjugated Secondary Antibody
  • AP-Conjugated Secondary Antibody
  • Substrate F1-specific for HRP
  • Substrate F2-specific for AP
  • Blocking Buffer
  • Wash Buffer
  • Plate Sealers

*Kit contents may vary slightly. Please refer to product-specific inserts for a complete list of contents for individual assays.

Advantages

  • No specialized equipment needed
  • No lengthy sample preparation
  • Results with as little as 10,000 cells/well
  • Measuring total and phospho-proteins simultaneously in the same well allows for a more accurate normalization of well-to-well variations.
  • Amenable to high-throughput screening of kinase inhibitors
  • Antibody specificity verified by Western blot
