For the detection of Phosphorylated Analytes by Sandwich ELISA

Antibodies specific for the phosphorylated and non-phosphorylated versions of the analyte (capture antibody; red) are pre-coated onto a microplate. The sample is added and any analyte (orange) present is bound by the immobilized antibody. An enzyme-linked specific for the phosphorylated version o fthe analyte detection antibody (blue) binds to a second epitope on the analyte forming the analyte-antibody complex.

Substrate is added and converted by the enzyme, thereby producing a colored product or light emission in direct proportion to the amount of analyte bound in the initial reaction (signal increases as concentration increases).
