CultreCoat® Cell Invasion Assays

CultreCoat Cell Invasion Assays offer a ready-to-use approach for measuring cell migration through extracellular matrices. These kits contain a 96-well Boyden chamber with the PET membrane pre-coated with Basement Membrane Extract (BME). The use of pre-coated plates improves reproducibility of results because the coating process assures uniform layers of extracellular matrix plate-to-plate and lot-to-lot, minimizing variability. Since cell lines exhibit different invasive capacities, the CultreCoat BME Cell Invasion Assays are available in a low, medium or high matrix coating density. In addition, a CultreCoat BME Cell Invasion Optimization Assay is available for determining the ideal coating density for your study. The optimization assay includes one 96-well Boyden chamber with the PET membrane pre-coated with three different concentrations of BME and uncoated wells to compare invasion to basal migration. Once it is determined which concentration provides the maximum signal-to-noise ratio, low, medium, or high density BME-coated plates can be ordered.

Quantification of Cell Invasion through Different BME Densities

FBS Stimulates Migration of MDA-MB-231 and HT1080 Cells. The MDA-MB-231 human breast cancer cell line (cyan bars) and the HT1080 human fibrosarcoma cell line (magenta bars) were treated with 10% fetal bovine serum (FBS). Migration of MDA-MB-231 and HT1080 cells through different BME densities was measured using the CultreCoat BME Cell Invasion Optimization Assay (Catalog # 3484-096-K). Data from four experiments was quantified for both cell types.
