Dual-Color ELISpot Kits
R&D Systems' Dual-Color ELISpot Kits are designed for the simultaneous quantitation of the frequency of cells secreting either one or both of two cytokines at the single cell level. Complete ELISpot kits are ready-to-run and require no assay development or refinement.
Dual-Color ELISpot assays are used to measure responses with frequencies well below 1 in 100,000 cells. This technique does not require prior in vitro expansion of cells and is suitable for high throughput analysis using only small numbers of primary cells. As such, ELISpot assays are useful tools for research in vaccine development. However, the secretion of one cytokine alone often does not correlate with long-term immune responses. For example, IFN-gamma secretion is often used to assess immune response to pathogens. It is typically associated with the Th1 response and is linked to the cytotoxic capacity of T cells. However, in some instances, measurement of IFN-gamma alone does not correlate with long-term immunity or pathogen control. When paired with the simultaneous measurement of IL-2, vaccine testing on malaria-exposed individuals indicates that dual-secreting cells were a much better indicator of resting memory after three months than either IFN-gamma or IL-2 alone.
