Sample Preparation
Total prepared sample volume needed per well is 50 microliters.
Serum and plasma samples require a 30-fold dilution. A suggested 30-fold dilution is 10 microliters of sample + 290 microliters of Calibrator Diluent RD5P. When assaying sVCAM-1, serum and plasma samples must be further diluted 4-fold to a final 60-fold dilution. Minimum sample volume recommended for sample preparation is 10 microliters. The minimum starting serum/plasma volume recommended for sample dilution is 10 microliters. While 50 microliters of prepared (diluted) sample is added to each well, the actual serum/plasma volume in each well will be less than 10 microliters. Pipetting less than 10 microliters of serum/plasma is not recommended due to potential pipetting inaccuracy. Samples may be further diluted if necessary to bring sample data within the standard curve range.
Cell culture supernates can be run undiluted with no matrix effects or interference. Maximum sample requirement per well is 50 microliters. Samples may be further diluted if necessary to bring sample data within the standard curve range.
