* A standard curve must be generated each time an assay is run, utilizing values from the Standard Value Card included in the Base Kit.
** CD40 Ligand is present in platelet granules and is released upon platelet activation. Therefore, to measure circulating levels of CD40 Ligand, platelet-free plasma should be collected for measurement (as described in the Product Insert). It should be noted that many protocols for plasma preparation, including procedures recommended by the Clinical and Laboratory Standards Institute (CLSI), result in incomplete removal of platelets from blood. This will cause variable and irreproducible results for assays of factors contained in platelets and released by platelet activation. |
Sample Preparation
Total prepared sample volume needed per well is 50 microliters.
Serum and plasma samples require a 10-fold dilution. For testing samples in duplicate, a suggested 10-fold dilution is 20 microliters of sample + 180 microliters of Calibrator Diluent RD6-56.
