Analysis of IL-1beta-induced COX-2 Expression using the COX-2 Cell-Based ELISA. CCD-1070SK human foreskin fibroblasts were cultured in 96-well microplates and treated with the indicated concentrations of Recombinant Human IL-1 beta (Catalog # 201-LB) for 24 hours. COX-2 levels were determined using the Human/Mouse Total COX-2 Cell-Based ELISA (Catalog # KCB4198) and normalized to total GAPDH detected in the same microplate wells (bar graph). Western blot analysis using the antibodies provided in the kit is shown for comparison (inset).

This fluorescence-based assay simultaneously measures COX-2 and GAPDH in the same microplate well without the need for lysate preparation. Normalization of the fluorescent signal derived from COX-2 to that of the GAPDH protein corrects for well-to-well variations such as differences in cell number. This Cell-Based ELISA Kit includes primary antibodies to COX-2 and GAPDH, species-specific secondary antibodies conjugated to either horseradish peroxidase (HRP) or alkaline phosphatase (AP), spectrally distinct fluorogenic substrates for each, and wash and blocking buffers. Cell-Based ELISAs require no specialized equipment and can be run with as few as 10,000 cells per well, making them an excellent alternative to Western blot.
