Application: Western BlotSample Tested: Human pancreatic cancer cell lines MiaPaCa-2, AsPC-1, and BxPC-3Species: HumanVerified Customer | Posted 03/20/2019Lane 1-AAVS1 guide RNA Lane 2-CLNS1A/pICLn targeting guide RNAMiaPaca2 cells stably expressing Cas9 were infected with AAVS1 control or CLNS1A/pICLn targeting guide RNA lentivirus for 48h, selected for guide RNA infection with 1ug/ml puromycin for an additional 48h. Cells were lysed in RIPA, cleared by centrifugation, soluble protein was quantified by BCA, and 20ug of whole cell extract was added in each lane. CLNS1A/pICLn was detected with this antibody at 1:2,000 dilution overnight at 4C followed by secondary antibody 1:5,000 (LiCor) for 30 minutes at room temp and developed on a LiCor imager.
