Detects mouse and rat DARPP-32 when phosphorylated at T34 in Western blots. There is weak cross-reactivity with phosphorylated PP1 Inhibitor-1, which contains a nearly identical phosphorylation site. Reactivity with human Phospho-DARPP-32 has not been confirmed.

Product Feature: CoraFluor Probes for TR-FRET

CoraFluor™ 1, amine reactive (Catalog:7920) and CoraFluor™ 2, amine reactive (Catalog # 7950) are terbium-based probes that have been developed for use as TR-FRET donors. They emit wavelengths compatible with commonly used fluorescent acceptor dyes such as BODIPY® (or BDY) and Janelia Fluor® dyes, FITC (Catalog # 5440), TMR and Cyanine 5 (Catalog # 5436). CoraFluor™ fluorescence is brighter and more stable in biological media than existing TR-FRET donors, leading to enhanced sensitivity and improved data generation. CoraFluor™ 1 exhibits excitation upon exposure to a 337 nm UV laser.

Background: DARPP-32

DARPP-32, a dopamine (DA) and cAMP-regulated ~32k phosphoprotein, is associated with dopaminoceptive neurons bearing D-1 receptors in the basal ganglia. The protein inhibits protein phosphatase I when it is phosphorylated on Thr34. In contrast, when DARPP-32 is phosphorylated on Thr75 the protein acts as an inhibitor of PKA. Phosphorylation of DARPP is thought to play a critical role in the regulation of dopaminergic neurotransmission. In addition, the activity of DARPP-32 is also thought to play important roles in the actions of alcohol, caffeine and Prozac
