Scientific Data Images for FKBP51 Antibody
FKBP51 in Human Prostate.
FKBP51 was detected in immersion fixed paraffin-embedded sections of human prostate using 15 µg/mL Human/Mouse/Rat FKBP51 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4094) overnight at 4 °C. Tissue was stained with the Anti-Goat HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS008) and counterstained with hematoxylin (blue). View our protocol for Chromogenic IHC Staining of Paraffin-embedded Tissue Sections.Detection of Human/Mouse/Rat FKBP51 by Western Blot.
Western blot shows lysates of DU145 human prostate carcinoma cell line. PVDF membrane was probed with 0.3 µg/mL of Human/Mouse/Rat FKBP51 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4094) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). A specific band was detected for FKBP51 at approximately 51 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 2.Detection of Human FKBP51 by Simple WesternTM.
Simple Western lane view shows lysates of DU145 human prostate carcinoma cell line, loaded at 0.2 mg/mL. A specific band was detected for FKBP51 at approximately 61 kDa (as indicated) using 3 µg/mL of Goat Anti-Human/Mouse/Rat FKBP51 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4094) followed by 1:50 dilution of HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.Western Blot Shows Human FKBP51 Specificity by Using Knockout Cell Line.
Western blot shows lysates of HeLa human cervical epithelial carcinoma parental cell line and FKBP51 knockout HeLa cell line (KO). PVDF membrane was probed with 0.3 µg/mL of Goat Anti-Human/Mouse/Rat FKBP51 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4094) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for FKBP51 at approximately 32 kDa (as indicated) in the parental HeLa cell line, but is not detectable in knockout HeLa cell line. GAPDH (Catalog # AF5718) is shown as a loading control. This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.Detection of FKBP51 by Immunohistochemistry
A limited number of genes are uniquely expressed in the GF small intestine. (A) Venn diagrams comparing gene expression profiles in small intestinal segments of CR, N, and O animals when contrasted with the gene expression profile obtained in GF mice. Eleven gene probes (representing 10 annotated genes) and 9 gene probes (representing 8 annotated genes) were differentially expressed in the SI-middle or SI-distal segment, respectively (selection criterion: |log FC| ≥ 1; p < 0.05 in at least two of three comparisons). The genes are listed in Supplementary Table S3. (B) Validation of cDNA microarray by qRT-PCR. Diagrams depict expression analysis of the indicated genes performed by cDNA microarray hybridization (left) or by qRT-PCR (right). Normalized fluorescence signal obtained in the respective gene probe upon hybridization with RNA isolated from GF mice was set to 1. Results of qRT-PCR were normalized to ubiquitin B (Ubb); the expression level of the respective gene in the mucosa of GF mice was set to 1. Four mice from each group were analyzed. Error bars represent SDs. Corresponding CT values are listed in Supplementary Table S4; * p < 0.05; ** p < 0.01 [one-way Analysis of Variance (ANOVA) test]. (C) Immunohistochemical detection of Fkbp5 and Irs2 using 3,3’-diaminobenzidine (DAB) staining (brownish precipitate) in the SI-middle part using specimens obtained from animals of the indicated experimental group. Both proteins are mainly produced in epithelial cells (black arrows). In addition, prominent Irs2 staining was also observed in the smooth muscle layer (green arrows). Representative images of specimens counterstained with hematoxylin are shown. Scale bar: 0.15 mm. Image collected and cropped by CiteAb from the following open publication (https://pubmed.ncbi.nlm.nih.gov/30934845), licensed under a CC-BY license.
