FRS2 (FGF R substrate 2; also known as SNT and FRS2 alpha ) is a 70‑90 kDa member of the FRS family of lipid-anchored docking proteins. It is an intermediary between FGF R and other RTK receptors and their Ras/MAPK signaling cascades. FRS2 contains a membrane-anchoring myristoylation signal (aa 1‑6), a PTB domain (aa 13‑115) that interacts with FGF and NGF receptors, and a C-terminal Tyr-rich region (aa 196‑471) that serves as a docking site for GRB‑2 and SHP‑2.

Detection of Human/Mouse/Rat FRS2 by Western Blot.
Western blot shows lysates of MCF-7 human breast cancer cell line, C2C12 mouse myoblast cell line, and PC-12 rat adrenal pheochromocytoma cell line. PVDF membrane was probed with 1 µg/mL of Goat Anti-Human/Mouse/Rat FRS2 Monoclonal Antibody (Catalog # MAB4069) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for FRS2 at approximately 85 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.Detection of FRS2 by Western Blot
The association between circKCNN2 and the therapeutic effect of lenvatinib. (A) The proliferation of Huh7 and HepG2 cells were inhibited by different doses of lenvatinib. (B) The effect of lenvatinib treatment and circKCNN2 overexpression on the proliferation of Huh7 cells. (C) The effect of lenvatinib treatment and circKCNN2 overexpression on the apoptosis of Huh7 cells. (D) Lenvatinib treatment elevated the RNA level of circKCNN2 in Huh7 cells. The DMSO group served as a reference for normalization. (E) circKCNN2 and lenvatinib enhanced the expression of MBD2 in Huh7 cells. The group without treatment served as a reference for normalization. (F) The effect of circKCNN2 and lenvatinib on the protein levels of FRS2, FGF19, and FGFR4 in Huh7 and HepG2 cells. (G) The miR‐520c‐3p mimics and the knockdown of MBD2 can reverse the circKCNN2‐induced downregulation of FGFR4. (H) The inherent levels of circKCNN2 in the two HCC organoids. Org543 served as a reference for normalization. (I) The effect of circKCNN2 overexpression on the cell growth of HCC organoids. (J) The effect of lenvatinib on the cell growth of HCC organoids. All the assays were performed at least three times. n = 3 for each group. *P < 0.05, **P < 0.01, ***P < 0.001 Image collected and cropped by CiteAb from the following open publication (https://pubmed.ncbi.nlm.nih.gov/35051313), licensed under a CC-BY license.
