Commercially available antibodies often contain substances (e.g. BSA, glycine, tris, azide) that interfere in labeling reactions. The AbSelect Purification Kit quickly removes these contaminants. It can also be used to purify antibodies from crude samples such as ascites fluid or immune serum.

The antibody to be purified or cleaned up ideally is in a volume of 100 ul to 0.5 ml. Up to 500 ug of antibody can be purified in each run. The AbSelect method involves capture of the antibody on protein A resin and the removal of unwanted substances by a simple wash procedure. The purified product is then eluted and neutralized.

The components of the AbSelect kit are fully compatible with the Lightning-Link antibody labeling system (available separately), which allows the purified antibody to be rapidly conjugated with a wide variety of labels.

Application Notes

The volume of antibody to be purified or cleaned up should be 0.1-0.5 mL, though larger volumes may be processed by first incubating the antibody sample (combined with the Binding Buffer) with the Protein A resin in a larger vessel (e.g. 2 mL-eppendorf tube) prior to transferring to the spin cartridge in several aliquots, spinning down the excess liquid each time.

  • Q: Can AbSelect Antibody Purification Kits be used for purification of other proteins in addition to antibodies?

    A:

    No, the AbSelect Antibody Purification Kit is not suitable for purifying proteins other than antibodies. This kit uses a Protein A resin. Protein A binds to antibodies but not other proteins. The antibody molecules are therefore bound to the resin, while any other proteins in the sample are simply washed through in the wash step, and hence are removed. The antibody is then detached from the resin in the elution step. Therefore, you should not attempt to purify anything other than antibodies using an AbSelect kit. However, you may be interested in one of our Antibody Concentration and Clean Up Kits, catalog numbers 861-0010 (3 columns) and 861-0005 (1 column). This product, in contrast to AbSelect, purifies by size: the sample is passed through a filter with a 10 kDa molecular weight cut-off. Therefore, if your protein is not too small, and you only want to remove low molecular weight substances, you can use the Antibody Concentration and Clean Up Kit.
