What is Simple Plex Technology?  

Simple Plex Technology combines the Ella Automated Immunoassay System, with ready-to-run Simple Plex Assays for unmatched precision and consistency. Together, they deliver fast, hands-free results you can rely on—with the ability to measure up to eight analytes in less than 90 minutes.

Simple Plex Immunoassays leverage microfluidic cartridges to obtain high-sensitivity biomarker detection, combining precision and efficiency within a closed, automated platform. This system relies on microfluidic technology to miniaturize and streamline traditional immunoassay workflows, allowing for multiplexed analysis with minimal sample volume. By incorporating pre-loaded reagents and automated handling, Simple Plex immunoassays offer reduced variability and faster results compared to conventional methods like ELISA.

How does it work?

  • Sandwich immunoassay occurs within Glass Nano Reactors (GNRs) 
  • 2-3 GNRs within each channel provide duplicate/triplicate results per sample 

Microfluidic Sandwich ELISA

  • Sample is routed through a microfluidic circuitry, with synchronized release of assay reagents 
  • Cartridges are pneumatically operated, pumping liquids from different reservoirs throughout the assay run

Single Analyte Immunoassays

Single Plex Assays allow you to quantify 1 analyte in a single microfluidic channel. Single Plex Simple Plex cartridges are available in 16, 32, and 72 sample configurations, with built-in triplicate measurements. When you need high-quality data from your sample and high throughput, the 72x1 cartridge offers a fast and scalable option. 

Multi-analyte Immunoassays

Multianalyte assays allow you to quantify 2, 3, or 4 target analytes in parallel. Each microfluidic channel has a unique immunoassay sandwich, consisting of high-quality capture and detection antibodies. Multianalyte Simple Plex cartridges are available in 16 and 32 sample configurations, with built-in triplicate measurements for every analyte. When you need high-quality data from your sample and high throughput, the 32x2, 32x3, and 32x4 cartridges offer flexible options.  

Multiplex Immunoassays

Multiplex assays allow you to quantify 5, 6, 7, or 8 analytes in duplicate. Each microfluidic channel contains two spatially separated capture antibodies so you can quantify up to 8 target analytes per sample. This cost-effective assay allows you to maximize the data that you can extract from each of 32 samples in 90 minutes or less. Generate up to 256 reportable results from each multiplex assay.

Using fluorescent readout, Simple Plex immunoassays on Ella delivers better sensitivity compared to traditional colorimetric ELISA kits allowing for sensitive detection and up to 4+ logs of dynamic range.

The 4-log dynamic range enables accurate quantification of a wide range of analyte concentrations within a single run, minimizing the need for repeat measurements with multiple dilutions. This also reduces overall sample volume requirements.

Fully Validated Immunoassay Solution  

Simple Plex Assay validation criteria to help ensure robust, reproducible, and validated results.  

Linearity- is a measure of accuracy with diluted samples. In Simple Plex Assays, all sample types are optimized for linear dilutions through the standard curve range. 
Specificity- or true negative rate, is confidence in a negative test result. Immunoassays are subject to several potential interferences from components in complex samples like serum or plasma. Simple Plex Assays are rigorously tested to ensure the correct intended analyte is measured.
Recovery- or spike recovery, is a measurement of accuracy. A spike recovery study provides an estimate of how much of the available analyte is measurable in the assay and if there may be sample components interfering with that measurement. Simple Plex Assays are optimized for recovery at three different spike concentrations (low, mid, high) and for every validated sample type. 
Precision- Consistency, or reproducibility, of a method measures precision. Simple Plex Assays ensure lot-to-lot consistency, enabling long-term research studies. This, combined with our stringent manufacturing and quality control measures, helps ensures reliable and reproducible results between kit lots and prevents sample value drift.
