FAQs for pCL-Eco Retrovirus Packaging Vector

Q: Can I use your packaging plasmid PCL-Eco Retrovirus packaging vector (pCLNDX Retrovirus expression Vector [NBP2-29540]) with lentiviral vector? I have been using the system of plasmids carrying the gag-pol, rev, GFP-lentivirus and vsv-g (to infect human cells). Can I use the PCL-Eco to infect mouse cells instead of vsv-g?
A: Unfortunately, we have not tested lenti-virus in this packaging kit so I cannot tell you if it will work or not. There are lots of packaging kits on the market for lenti-virus so if you take a look maybe you can find one that will be certain to work. We, however, do not have a lenti-virus kit.

Q: I am having a retroviral system in place which allows me to infect mouse cell lines. The vectors I have are MIG.R (with my gene of interest), pCL-ECO and VSV-g. I know that my pCL-ECO vector is only suitable for infecting mouse cells. My question is: Is it possible to use my MIG.R and VSV-g vectors, and exchange the mouse specific pCL-ECO with pCL-Ampho or pCL-10A1 packaging vector which you both offer for purchase (Cat Numbers are 10045P, 10046P, 10047P). My aim is to infect human cell lines with my gene of interest.
A: These vectors, while also sold separately, are components of the RetroMax system 10040K. The vectors are designed to be used as described in the 10040K manual. Yes, you are correct, pCL-ECO is for mouse and rat, and not for human. We have not evaluated the Retromax system with the MIG.R and VSV-g vectors, and can not promise they would be compatible with your vectors. The Retromax packaging vectors are designed to be used with the expression vectors in the Retromax Kit. However, the packaging vectors are widely published and you may be able to find some information in the literature relevant to your model system (search scholar.google.com and keywords with Retromax and vector name, or keys words with packaging vector name and expression vector name ).

Q: I'm currently producing a retrovirus with the pCL-Eco packaging vector. However, my target cells are not getting infected, so I was thinking of switching to a lentivirus. Can I use this pCL-Eco packaging vector with a lentivirus, and if so, is it still only infectious to rodents?
A: Unfortunately, we have not yet tested the pCL-Eco vector kit with lentivirus, therefore, we would not be able to provide you with any information regarding this type of vector. I am sorry for any inconvenience.

Q: What is the packaging size limitation on Eco Retrovirus Packaging Vector NBP2-29540?
A: The combined size of two inserts should not be more than 4 kb.
