Samples: Whole cell lysate (50 ug) from HeLa, HEK293T, Jurkat, and mouse TCMK-1 cells prepared using NETN lysis buffer. Antibody: Affinity purified rabbit anti-AF6 antibody NBP1-22996 used for WB at 0.1 ug/ml. Detection: Chemiluminescence with an exposure time of 10 seconds.

Afadin/AF-6 Antibody [NBP1-22996] - Detection of human AF6 by western blot of immunoprecipitates. Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from HeLa cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-AF6 antibody NBP1-22996 (lot NBP1-22996-2) used for IP at 6 ug per reaction. AF6 was also immunoprecipitated by a previous lot of this antibody (lot NBP1-22996-1) and a another rabbit anti-AF6 antibody. For blotting immunoprecipitated AF6, NBP1-22996 was used at 1 ug/ml. Detection: Chemiluminescence with an exposure time of 30 seconds.

Background: Afadin/AF-6

AF6 was identified as part of a fusion with MLL (mixed lineage leukemia) in a t(6;11) genetic translocation. AF6 is also known as afadin (a splicing variant of AF6 found at adherins junction). AF6/afadin is an actin filament binding protein that binds to nectin at cadherin-based cell-cell adherens junctions and regulates cell-cell adhesion by mediating Ras signaling.
