Western Blot:[NBP3-30031]-Detection of human CDC6 by western blot and immunoprecipitation. Samples: Whole cell lysate from HeLa and HEK293T cells. Antibodies: Affinity purified rabbit anti-CDC6 antibody used for WB at 0. 4 µg/ml and 1 µg/ml and used for IP at 3 µg/mg lysate. CDC6 was also immunoprecipitated by rabbit anti-CDC6 antibody, which recognizes an upstream epitope. Detection: Chemiluminescence with exposure times of 3 minutes.

Background: Cdc6

Cdc6 is encoded by this gene is highly similar to Saccharomyces cerevisiae Cdc6, a protein essential for the initiation of DNA replication. This protein functions as a regulator at the early steps of DNA replication. It localizes in cell nucleus during cell cyle G1, but translocates to the cytoplasm at the start of S phase. The subcellular translocation of this protein during cell cyle is regulated through its phosphorylation by Cdks. Transcription of this protein was reported to be regulated in response to mitogenic signals through transcriptional control mechanism involving E2F proteins.
