Immunoprecipitation:BAP28 AntibodyNBP3-29875] -Detection of human HEATR1 by western blot of immunoprecipitates. Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from 293T cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-HEATR1 antibody NBP3-29875 (lot NBP3-29875-1) used for IP at 6 µg per reaction. HEATR1 was also immunoprecipitated by rabbit anti-HEATR1 antibody BL19992. For blotting immunoprecipitated HEATR1, NBP3-29875 was used at 0.4 µg/ml. Detection: Chemiluminescence with an exposure time of 3 seconds.

Background: BAP28

IRAK-4 is a member of the interleukine-1 receptor-associated kinase (IRAK) family. The IRAK family is implicated in the Toll-like receptor (TLR) and Il-1R signaling pathway (1). Upon phosphorylation, IRAK-4 interacts with IRAK-1, MYD88 and TRAF6 to form a complex, and activate NF-kB and MAPK pathways (2-3). IRAK-4 phosphorylates and activates IRAK-1 at thr387 and is also known to interact with IRAK-1 but not with the other IRAK members. Additionally, like IRAK-1, IRAK-4 possesses an auto and cross-phosphorylation kinase activity (4).
