Western blot shows lysates of Balb/3T3 mouse embryonic fibroblast cell line and CH-1 mouse B cell lymphoma cell line. PVDF membrane was probed with 1 µg/mL of Mouse MGMT Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3299) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF017). A specific band was detected for MGMT at approximately 25 kDa (as indicated). This experiment was conducted using Immunoblot Buffer Group 1.

Background: MGMT

The O-6-methylguanine-DNA-methyl transferase (MGMT) is involved in the direct repair of DNA alkylated at the O6 position of guanine. MGMT transfers the alkyl group to its own cysteine thereby inactivating the MGMT molecule. Therefore, the number of MGMT molecules correlates directly with the cell’s ability to withstand DNA alkylation.
