Western Blot: PHLPP2 Antibody [NB100-1812] - Whole cell lysate (50 ug) from HeLa, RKO, MCF-7, TCMK-1, and NIH 3T3 cells prepared using NETN lysis buffer. Antibody: Affinity purified Rabbit anti-PHLPPL antibody used for WB at 0.04 ug/ml. Detection: Chemiluminescence with an exposure time of 75 seconds.
PHLPP2 Antibody [NB100-1812] - Detection of human PHLPPL by western blot of immunoprecipitates. Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from HeLa cells prepared using NETN lysis buffer. Antibodies: Affinity purified Rabbit anti-PHLPPL antibody NB100-1812 (lot NB100-1812-2) used for IP at 6 ug per reaction. PHLPPL was also immunoprecipitated by a previous lot of this antibody (lot NB100-1812-1). For blotting immunoprecipitated PHLPPL, NB100-1812 was used at 0.04 ug/ml. Chemiluminescence with an exposure time of 10 seconds.

Background: PHLPP2

PHLPPL (PH domain leucine-rich repeat-containing protein phosphatase-like) is responsible for the dephosphorylation and subsequent inactivation of Akt. Akt is a signaling protein critical to the balance of cell survival and apoptosis. Dephosphorylation of Akt by PHLPPL has been demonstrated to trigger apoptosis and suppress tumor cell growth. PHLPPL is also known as PHLPPL2 and functions similarly to its relative, PHLPP1. Recent studies show that PHLPPL and PHLPP selectively terminate Akt-signaling by targeting specific Akt isoforms. Alternate names for PHLPPL include PH Domain and Leucine Rich Repeat Protein Phosphatase-Like, KIAA0931, and PHLPP2.
