RiboDyEE™: A New Cost-Effective Solution for Robust RNA Encapsulation Efficiency Measurement in Lipid Nanoparticles
Application NotesRNA-based therapeutics have gained considerable attention in the field of biopharmaceutics with lipid nanoparticles (LNPs) serving as an essential delivery vehicle. RNA encapsulation efficiency (EE) measurement within LNPs is critical to ensure dosing accuracy and efficiency and is commonly measured using a fluorescence-based assays with a single fluorescent dye. Although such assays are easy to perform in standard laboratory setups, they lead to inaccuracies when measuring LNP EE.
Recognizing the need to promote best practices for consistent LNP EE assays, we have developed RiboDyEE, the first commercially available dye specifically for measuring LNP EE.
This application note describes the validation of RiboDyEE in a new encapsulation assay format for mRNA-LNP EE determination and gives tips for best usage practices.
