Scalable Manufacturing of Functional Beta Islets

Human induced pluripotent stem cells (hiPSCs) offer a powerful platform for regenerative medicine due to their ability to self-renew and differentiate into virtually any cell type. In type 1 diabetes, the loss of insulin-producing pancreatic beta cells has driven significant interest in generating functional pancreatic islets from hiPSCs as a restorative cell therapy. However, advancing these therapies toward clinical and commercial implementation requires scalable manufacturing workflows, GMP-grade reagents, and robust analytical tools to support process consistency and product characterization.

In this application note, we demonstrate a scalable 2D-3D workflow for generating pancreatic islet-like clusters from hiPSCs using R&D Systems™ Animal-Free GMP-grade reagents and PBS Biotech Vertical-Wheel® bioreactors. By combining a reproducible differentiation process with comprehensive cell characterization, this integrated workflow supports the production and evaluation of hiPSC-derived functional pancreatic islets for diabetes research and translational cell therapy applications.

Key Takeaways

  • Vertical-Wheel bioreactors provide a scalable and reproducible platform for generating hiPSC-derived pancreatic islet-like clusters, supporting the translation of stem cell differentiation workflows into manufacturable cell therapies.
  • R&D Systems Animal-free, GMP-grade proteins and small molecules supported robust cell growth, lineage specification, and maturation throughout the differentiation workflow, reinforcing translational readiness.
  • A combination of R&D Systems antibody-based cell characterization tools and Simple Plex™ Assays on the Ella™ System enabled efficient assessment of cell identity, maturation, and glucose-responsive insulin secretion.
  • Together, this integrated manufacturing and analytical workflow supports the scalable production and evaluation of hiPSC-derived pancreatic islets, helping accelerate the development of stem cell-based therapies for type 1 diabetes.
