Phospho-p130Cas/BCAR1 (Tyr-762) synthetic peptide (coupled to KLH) corresponding to amino acid residues around tyrosine 762 in the Src-binding domain of rat p130Cas/BCAR1. This rat sequence is the same as Tyr-664 in human p130Cas/BCAR1 and Tyr-668 in mouse p130Cas/BCAR1. This site is also highly conserved in Cas-L (HEF1, Nedd9). [UniProt# Q63767]

Specificity

This antibody was cross-adsorbed to phospho-tyrosine coupled to agarose and to phospho-p130Cas/BCAR1 (Tyr-751) peptide before affinity purification using phospho-p130 Cas (Tyr-762) peptide (without carrier). The antibody detects a 130 kDa protein corresponding to the molecular mass of phosphorylated p130 Cas on SDS-PAGE immunoblots of pervanadate treated human A431 cells, but not in control cells.

Background: p130Cas

BCAR1/p130CAS functions as a scaffolding protein for components of various kinase signaling pathways. It is involved in integrin signaling for cell motility as well as netrin signaling for the guidance of commissural axons. Overexpression of BCAR1/p130CAS has been shown to confer antiestrogen resistance in breast cancer cells. Alternate names for BCAR1/p130CAS include breast cancer anti-estrogen resistance protein 1, CRK-associated substrate, CAS scaffolding protein family member 1, CAS, and CRKAS.
