To resolve the challenges of purifying hydrophobic Wnt proteins in an active state and using multiple proteins in a single experiment, R&D Systems™ has engineered a Wnt agonist protein that can act as a Wnt and R-Spondin pathway activator to promote canonical Wnt signaling. As seen in this application note, Wnt/R-Spondin Agonist Proteins display better activity in a Wnt/β-Catenin assay compared to Wnt-3a with or without R-Spondin proteins. Wnt/R-Spondin Agonist Proteins are ideal for organoids and 3D cell culture, as they support robust intestinal organoid culture at lower concentrations relative to Wnt-3a and R-Spondin 1 together.
