Specificity

Recognizes human Lymphotoxin alpha chains in alpha 2 beta 1 or alpha 1 beta 2 trimers and also human TNF-beta. In direct ELISAs, less than 10% cross-reactivity with recombinant human (rh) APRIL, rhBAFF, rhEDA-A2, rhFas Ligand, rhLIGHT, rhGITR Ligand, rhOX40 Ligand, recombinant mouse RANKL, rhTNF-alpha, rhTRAIL, rhTRANCE, rhTWEAK, or rhVEGI is observed.

Mouse Anti-Human Lymphotoxin Antibody Clone # 135125 (Catalog # MAB1370) neutralizes Recombinant Human Lymphotoxin alpha 2/ beta 1 (679-TX) induced cytotoxicity in the L-929 mouse fibroblast cell line in the presence of actinomycin D. The ND50 for this effect is typically 0.150-3.00 µg/mL.

Neutralization

Measured by its ability to neutralize Lymphotoxin alpha 2/ beta 1­induced cytotoxicity in the L­929 mouse fibroblast cell line. The Neutralization Dose (ND50) is typically 0.150 - 3.00 µg/mL in the presence of 10 ng/mL Recombinant Human Lymphotoxin alpha 2/ beta 1and actinomycin D.

Background: Lymphotoxin

Lymphotoxin alpha, also known as tumor necrosis factor (TNF) beta, and Lymphotoxin beta are members of the TNF superfamily and have been designated TNFSF1 and TNFSF3, respectively. Secreted Lymphotoxin alpha forms a homotrimer (TNF­beta) and heterotrimeric complexes with the type II membrane protein, Lymphotoxin beta (either alpha 2 beta 1or alpha 1 beta 2 timers). These trimers serve as ligands for Lymphotoxin beta receptor, TNF RI and TNF RII and play a role in the development and maturation of the immune system.
