Expression of RNF31 in HCC cell lines and evaluation of RNF31 knockdown. Inhibition of RNF31 with HOIPIN-8 decreases proliferation, invasion, and NF-kappa B activation in HCC cell lines. (a) Cell proliferation was evaluated in HepG2 and Hep3B cells treated with 100 uM HOIPIN-8. (b) Invasion was evaluated in HepG2 and Hep3B cells in the presence of 10 or 30 uM HOIPIN-8. Representative images of invading cells are shown in the right panel. Cell nuclei are stained in purple. (c) Expression of NF-kappa B target genes in HepG2 and Hep3B cells was evaluated by qPCR. Cells were pretreated with 30 uM HOIPIN-8 for 1 h, followed by stimulation with 10 ng/mL TNF-alpha for 2 h. (d) Phosphorylation of NF-kappa B signaling factors upon TNF-alpha stimulation was evaluated in HepG2 cells with or without 30 uM HOIPIN-8. Cell lysates were immunoblotted by the indicated antibodies. (e) Effect of HOIPIN-8 on TNF-alpha + CHX-mediated apoptosis in HepG2 and Hep3B cells. RNF31 knockdown inhibits TNF-alpha -induced NF-kappa B activation in HCC cell lines.
