Integrating TcBuster and Ambr with Multi-Attribute Characterization

This application note presents a streamlined workflow for developing and evaluating mAb-producing CHO cell lines using TcBuster™-mediated integration. High-throughput screening with the Ambr® 250 system enabled rapid optimization of culture conditions, while icIEF and CE-SDS on the Maurice System assessed key quality attributes such as purity and heterogeneity. Automated HCP analysis on the Ella™ System further enhances analytical efficiency.

Rapid Analysis for High-Throughput Antibody Development

This application note describes a workflow for recombinant antibodies that were produced on a HT platform, purified, and then analyzed for charge heterogeneity with icIEF and CE-SDS. Both methods resolved distinct peaks in all antibodies, signifying important structural properties that could potentially influence the selection process.

Assessment of Product Quality in Upstream Process Development

This whitepaper demonstrates the utilization of the Maurice System for the analysis of purity (CE-SDS) and charge heterogeneity (icIEF) of cell culture samples, both supernatants as well as purified antibodies. Turbo CE-SDS provided quantitative purity data and icIEF was able to detect and quantify small acidic and basic heterogeneity changes.

Accelerating Lead Identification Through Integrated Antibody Development and Characterization

This webinar demonstrates a scalable workflow to express and purify ~100 recombinant antibodies in a single run, an improved approach to CHO‑K1 cell line development using the TcBuster® transposon system, delivering significantly higher antibody titers and more productive clones, and high‑resolution biophysical methods using icIEF and CE-SDS techniques to rapidly assess size, purity, and charge heterogeneity—critical quality attributes that enable confident lead selection early in development. Together, these strategies provide an integrated path from antibody discovery to developable, manufacturable candidates—faster and with greater confidence.

Addressing the challenges of size and purity analysis

Hear about the recent advancements in biotherapeutic size analysis and purity analysis using the Maurice CE‑SDS platform. Caroline Daniels (Shattuck Labs) will demonstrate how her team uses the Maurice system to separate molecules based on differences in N‑linked glycosylation. After comparing the CE‑SDS Plus and Turbo cartridges for highly glycosylated heterodimeric fusion proteins, the Turbo CE-SDS cartridge was selected for both upstream and downstream routine use due to faster run times with maintained resolution.

Combining icIEF-Fractionation & Mass Photometry

Discover a novel workflow that streamlines charge–size variant characterization for complex antibody modalities.  Utilizing the MauriceFlex™ System and Refeyn’s TwoMP mass photometer, the speakers demonstrate a fast, simple application that utilizes charge variant separation and fractionation followed by  downstream charge–size variant analysis to uncover differences between a bispecific antibody (Mosunetuzumab-axgb) and a research-grade biosimilar.

Technical Tips

Optimizing Upstream Characterization of Antibodies using Maurice Platforms

In her training series “Technical Tips from Monica Tyagi,” Dr. Tyagi discusses some simple but important steps for using the Maurice platform to demonstrate how upstream clone selection can be accelerated through optimized, integrated antibody development and characterization workflows.

From Your Peers

Since adopting the Maurice instrument, our workflows have become significantly more accurate, efficient, and reproducible. We replaced less precise methods, such as analytical CEX, with high-resolution charge-variant characterization and reliable pI determination. The integrated CE-SDS capability now provides robust, quantitative size and purity data, enabling us to meet regulatory requirements for release assays.”
