Integrating TcBuster and Ambr with Multi-Attribute Characterization
This application note presents a streamlined workflow for developing and evaluating mAb-producing CHO cell lines using TcBuster™-mediated integration. High-throughput screening with the Ambr® 250 system enabled rapid optimization of culture conditions, while icIEF and CE-SDS on the Maurice System assessed key quality attributes such as purity and heterogeneity. Automated HCP analysis on the Ella™ System further enhances analytical efficiency.

This application note describes a workflow for recombinant antibodies that were produced on a HT platform, purified, and then analyzed for charge heterogeneity with icIEF and CE-SDS. Caroline Daniels (Shattuck Labs) will demonstrate how her team uses the Maurice system to separate molecules based on differences in N‑linked glycosylation. After comparing the CE‑SDS Plus and Turbo cartridges for highly glycosylated heterodimeric fusion proteins, the Turbo CE-SDS cartridge was selected for both upstream and downstream routine use due to faster run times with maintained resolution.

In her training series “Technical Tips from Monica Tyagi,” Dr. Tyagi discusses some simple but important steps for using the Maurice platform to demonstrate how upstream clone selection can be accelerated through optimized, integrated antibody development and characterization workflows.
