    HEK293 cells were loaded with 1 µM cytosolic Ca2+ sensitive dye, Fura-2AM in normal tyrode solution for 30 min and excited alternately at 340 and 380 nm and fluorescence was captured at 510 nm. ER Ca2+ store was depleted with 100 µM carbachol (CCh) in nominally Ca2+ free tyrode solution, resulting in transient cytosolic Ca2+ peak.
