    I used H2DCFDA to measure the overproduction of ROS by HepG2 cells under hypoxic and hyperglycemic conditions. Cells were treated with 10 μM of H2DCFDA for 30 min at 37 0C and imaged under a fluorescent microscope.

    Try different dilutions to get the one best suited to you. We treated the cells for 30 minutes and that was enough to get robust results
