Detection of Phospho-Ubiquitin (S65) by Western Blot. Western blot shows lysates of Di-Ubiquitin (negative control), Phospho-Tetra-Ubiquitin (S65), Phospho-Ubiquitin (S65), K48 linked Phosopho-Tetra-Ubiquitin (S65), and K48 linked Ubiquitin 2-7 (negative control). PVDF membrane was probed with 2 µg/mL of Rabbit Anti-Human Phospho-Ubiquitin (S65) Monoclonal Antibody (Catalog # MAB9866, left panel) or 0.1 µg/mL of Rabbit Anti-Human Ubiquitin Monoclonal Antibody (Catalog # MAB8595, right panel), followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # HAF008). A specific band was detected for Phospho-Ubiquitin (S65) at approximately 10, 30, and 35 kDa (as indicated on the left panel). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: Ubiquitin

Ubiquitin is a 76 amino acid, highly conserved nuclear and cytoplasmic protein. It is found exclusively in eukaryotes, becomes covalently attached to substrate proteins by enzymes in the Ubiquitin-Proteosome Pathway (UPP) and has a major role in targeting cellular proteins for the ATP-dependent degradation by the 26S proteosome. Ubiquitination also affects proteosome-independent events such as protein localization, activity and function.
