Activity
The peptide substrate contains a highly fluorescent 7-methoxycoumarin group that is efficiently quenched by resonance energy transfer to the 2,4-dinitrophenyl group. It can be used to measure the activities of peptidases that are capable of cleaving an amide bond between the fluorescent group and the quencher group, causing an increase in fluorescence. The substrate contains the Swedish mutation of the amyloid precursor protein (APP) beta -secretase cleavage site. A K-R-R sequence is added for easy solubilization. It can be used to measure the activity of BACE-1 (R&D Systems, Catalog # 931-AS) and thimet oligopeptidase (Koike, H. et al., 1999, J. Biochem. 126:235).

What does CF mean?

CF stands for Carrier Free (CF). We typically add Bovine Serum Albumin (BSA) as a carrier protein to our recombinant proteins. Adding a carrier protein enhances protein stability, increases shelf-life, and allows the recombinant protein to be stored at a more dilute concentration. The carrier free version does not contain BSA.

What formulation is right for me?

In general, we advise purchasing the recombinant protein with BSA for use in cell or tissue culture, or as an ELISA standard. In contrast, the carrier free protein is recommended for applications, in which the presence of BSA could interfere.
