Reactivity Notes

Rat, mouse and guinea pig in brain, spinal cord and ileum. Customer feedback suggests that human reactivity is negative. Sheep reactivity reported in scientific literature (PMID: 19912479).

Localization

In immunohistochemical experiments staining of the neuronal cell bodies and fibers is seen and staining is believed to be localized to the plasma membrane.

Specificity

NB300-102 is specific to the NK-3 receptor. Specificity was verified by the localization of immunohistochemical staining as well as pre-absorption studies with NK-3 peptide. (1)

Background: NK3R/TACR3

Neurokinins belong to tachykinin family of signalling peptides and the best known human tachykinins are neurokinin B, (encoded by TAC3) and substance P (SP) as well as neurokinin A (NKA), (both encoded by the same TAC1 gene). Neurokinins are widely expressed in CNS, and exert effects on target cells through action at three closely related Gq-coupled neurokinin receptors: NK1R, NK2R, NK3R: encoded in humans by TACR1, TACR2 and TACR3 respectively. None of these neurokinin receptors are entirely specific for SP, NKA or NKB, however, they do show some degree of selectivity, and TACR3/NK3R (neurokinin B receptor or neuromedin-K receptor) behaves as cognate receptor for NKB in vivo. NK3R are predominantly expressed in neurons of PNS/CNS and in particular, in many of forebrain areas, such as frontal, parietal and cingulate cortices, and basal ganglia structures implicated in psychiatric disease states, and can modulate monoaminergic and amino acid neurotransmission within these structures. Neurokinin B and NK3R are also expressed in placenta, kidney, skeletal muscle and lung tissues, and had been implicated in diverse processes, including water homeostasis, pulmonary inflammation, cognition, and preeclampsia.

Procedure Guide for NB 300-102 Polyclonal Anti-Neurokinin-3 (NK-B) Receptor
Immunohistochemistry

1) Tissue of interest is dissected and post-fixed in PBS containing 4% formaldehyde or PBS containing 4% formaldehyde + 0.1% gluteraldehyde**, followed by fixation in PBS containing 20% sucrose, overnight at 4C. Until the tissue sinks.
2) Cut fixed sections at 15 mm or 40 mm using a cryostat or sliding microtome, depending on incubation technique [for cryosections use 15 mm and for free-floating sections use 40 mm].
3) If using culture cells, remove media and perform 2-3 washed in PBS (pH 7.4) -use same protocol without agitation.
4) Wash sections in PBS for 10 minutes, 3 times.
5) Block the tissue with 1% BSA / 1% goat serum / 0.3% triton X-100 in 0.1M PBS, for 1 hour at RT.
6) Dilute anti-NK-3 receptor (NB 300-102) in blocking buffer at 1:1000 for cryosections and 1:5000 for free-floating sections.
7) Incubate sections overnight at 4C.
8) Wash sections in PBS for 10 minutes, 3 times.
9) Dilute secondary antibody (1:400 ?? Alexa 568 [Rho] goat anti-rabbit, from Molecular Probes, Inc.) in blocking buffer and incubate sections for 2 hours at RT.
10) Wash sections in PB (not PBS) for 10 minutes, 3 times.
11) Mount coverslips to slides using Fluoromount-G (Southern Biotech Assoc., cat# 0100-01)

**Note: Mouse dorsal horn and cerebral cortex were used as positive control section.

*Tissues fixed in 4% paraformaldehyde + 0.1% gluteraldehyde are used for EM analysis

  • Q: I was wondering about the Rabbit polyclonal antibody to Neurokinin-3-receptor, catalog #NB300-102. I am wondering about its information on the immunogen. You state that, NB300-102 was raised against a synthetic peptide at the c-terminus of rat Nk-3 conjugated to bovine thyroglobulin. I was wondering if you could tell me exactly which amino acids that this does correspond to? I am interested in comparing it to the Rabbit anti-Nk3R used for western blot, catalog#R-103-100, which states that it corresponds to the aa:434-452 of the absolute c-terminal of rat Nk3R protein.

    A: The immunogen for this antibody is between amino acids 375 and 452 of rat NK3R. Any further information regarding the immunogen is considered proprietary.
