Q: I received the Piezo-1 blocking peptide this morning. In the past, I have blocked antibodies for IHC by pre-absorbing the antibody with a 5-fold excess of blocking peptide for 2 hours at 4 degrees C before diluting the blocked mixture and applying to the slides. Can you confirm that this protocol is what you would recommend for effective blocking, or otherwise, suggest appropriate conditions. I am also assuming from the data sheet you sent that you recommend to reconstitute the peptide in water.
A: The peptide you received is not lyophilized but in solution form and the protocol you mentioned seems great.

Q: We have a customer who purchased NBP1-78446 and NBP1-78446PEP (PO12-01725/PO12-01822). The customer uses the antibody at a dilution of 1:10,000 in IHC successfully, but would like to use the blocking peptide to verify their IHC results. The customer would like to receive the recommended blocking peptide protocol for NBP1-78446PEP - most important, which is the recommended concentration/fold relation to antibody the customer should use!
A: We have a general recommended protocol for blocking peptides, which should be useful for you. You will see that in point 2., we say 'add the blocking peptide solution at a 5-10:1 ratio to your antibody, i.e. if you added 1 ug of antibody, add 5-10 ug of blocking peptide'.
