Immunoprecipitation: Rabbit Polyclonal RNA Polymerase II/POLR2A Antibody [NBP2-32080] - Detection of human RNA Polymerase II by western blot of immunoprecipitates. Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from HeLa cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-RNA Polymerase II antibody (NBP2-32080 lot 2) used for IP at 6 ug per reaction. RNA Polymerase II was also immunoprecipitated by a previous lot of this antibody and a second antibody against a different epitope of RNA Polymerase II antibody. For blotting immunoprecipitated RNA Polymerase II, the second antibody was used at 1 µg/ml. Detection: Chemiluminescence with an exposure time of 75 seconds.

Recruitment of RNA processing factors to the elongation complex is coordinated by the phosphorylation of Serine-2 and Serine-5 residues within the C-terminal repeat (YSPTSPS) domain of the largest subunit of RNAPII.

    Hep3B cells were lysed in immunoprecipitation lysis buffer and incubated 20 min on ice. Following centrifugation at 18,000 x g at 4C, lysates were precleared with protein G-Sepharose for 2 h with rotation. 1 mg of lysate was incubated with rotation overnight at 4C with 2 ug of NBP2-32080 antibody, followed by incubation with protein G-Sepharose for 2 h at 4C and washes with IP lysis buffer 4x for 5 min at 4C. Immunoprecipitates were used for immunoblot assay. WB was performed using a 1:1000 dilution of RNA Pol II antibody in 1% BSA/PBS.
