Western Blot: Protein Disulfide Isomerase/P4HB Antibody [H00005034-B01P] - Absence of TRIC-B alters expression of collagen-interacting proteins.(A) Quantitative RT-PCR of fibroblast transcripts encoding collagen, modifying enzymes & chaperones. Proband cells (P1, P2, & P3) demonstrate decreased expression of COL1A1 & FKBP10 (FKBP65), & increased expression of PLOD1 (LH1) & HSPA5 (BiP/GRP78) versus control fibroblasts (C). (B) Decreased expression of PLOD2 (LH2), FKBP10 (FKBP65) & PPIB (CyPB) in P2 osteoblasts versus control osteoblasts (C). (C) Immunoblots for quantitation of steady-state protein levels of collagen modifying enzymes & chaperones in proband & control cells. Proband fibroblasts show increases in PDI, LH1, LH2 & BiP protein levels. CyPB & FKBP65, both collagen-interacting isomerases, are consistently decreased in proband fibroblasts & osteoblasts. *, p < 0.05; **, p < 0.01; ***, p < 0.001.
