Immunoprecipitation:COASY AntibodyNBP3-29017] -Detection of human COASY by western blot of immunoprecipitates. Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from Jurkat cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-COASY antibody NBP3-29017 (lot NBP3-29017-1) used for IP at 6 µg per reaction. COASY was also immunoprecipitated by rabbit anti-COASY antibody. For blotting immunoprecipitated COASY, NBP3-29017 was used at 1 µg/ml. Detection: Chemiluminescence with an exposure time of 3 minutes.

Background: COASY

Biosynthesis of coenzyme A (CoA) from pantothenic acid (vitamin B5) is an essential universal pathway in prokaryotes and eukaryotes. COASY is a bifunctional enzyme that catalyzes the 2 last steps in CoA synthesis. These activities are performed by 2 separate enzymes, phosphopantetheine adenylyltransferase (PPAT; EC 2.7.7.3) and dephospho-CoA kinase (DPCK; EC 2.7.1.24), in prokaryotes (Daugherty et al., 2002 [PubMed 11923312]).[supplied by OMIM]
