Background: DMC1

DMC1 (disrupted meiotic cDNA or LIM15) is a meiosis-specific gene that encodes a nuclear protein which is essential for meiotic homologous recombination, specifically in homologous strand assimilation, and MDC1 also participates in formation of synaptonemal complex as well as double-strand breaks repair at recombination hotspots. DMC1 is a member of RecA family and in eukaryotic cells; there are two RecA homologs, RAD51 and DMC1, which promote the search for homologous DNA sequences and DNA strand exchange resulting in the formation of joint molecules. RAD51 acts both in meiotic recombinations as well as during DNA repair in mitotically dividing cells, however, DMC1 is expressed only during meiosis only. DMC1 interacts with the MND1-PSMC3IP heterodimer and with BRCA2. Dmc1-/- mice are infertile due to decreased germ cells, failing to form synaptonemal complexes and arresting at the leptotene or zygotene stage in late meiosis I. In female mice lacking Dmc1, normal oogenesis is aborted in embryos, and germ cells are depleted in the adult ovary. The phenotypes of Dmc1-/- ovaries are similar to clinical characteristics found in women with premature ovarian failure (POF), however, mutations in the coding sequence of DMC1 were shown to not to associate with POF in women.

Procedure Guide for NB 100-181 Anti-hDMC-1

Western Blot
1. Proteins are separated on an SDS-PAGE gel.

2. Following the protein transfer, the membrane is blocked with 5% NFDM in TBS-T.

3. The membrane is then washed.

4. NB 100-181 [anti-hDMC] is diluted 1:1,000 in dilution buffer [5% NFDM in TBS-T] and incubated for 3 hours at room temperature, gently shaking.

5. The membrane is then washed.

6. Secondary antibody [HRP conjugated goat anti-mouse] is incubated for 45 minutes at room temperature, gently shaking.

7. Develop membrane.

NOTE: Mouse testis tissue extracts were used as a positive control for this antibody. MEF 161 and HeLa whole cell extracts were not shown to work in Western analysis.
