Detection of Aiolos/IKZF3 in Human PBMCs by Flow Cytometry. Human peripheral blood mononuclear cells (PBMCs) were stained with Mouse Anti-Human CD19 APC‑conjugated Monoclonal Antibody (Catalog # FAB4867A) and either (A) Mouse Anti-Human Aiolos/IKZF3 Alexa Fluor® 488‑conjugated Monoclonal Antibody (Catalog # IC8625G) or (B) Mouse IgG1 Alexa Fluor 488 Isotype Control (Catalog # IC002G). To facilitate intracellular staining, cells were fixed and permeabilized with FlowX FoxP3 Fixation & Permeabilization Buffer Kit (Catalog # FC012). IKAROS family zinc finger 3 (IKZF3), also known as Aiolos, is a hematopoietic-specific transcription factor involved in the regulation of lymphocyte development. This gene product interacts with IKZF family memebers as well as FoxP3, resulting in select gene silencing in Tregs. Over amino acids (aa) 1-54, human and mouse share 70% aa sequence identity.
