Daxx (Fas death domain-associated-xx; also death domain-associated protein 6) is a ubiquitously expressed, proapoptotic protein that belongs to the Daxx family of molecules. Although it has a predicted MW of 81 kDa, it runs anomously at 110 kDa in SDS-PAGE. Daxx is found in the nucleus where it represses transcription. In the cytoplasm, it binds to ASK1, where it mediates Fas-induced apoptosis. Human Daxx is 740 amino acids (aa) in length and contains three coiled-coil regions (aa 180-217; 358-399; 430-489), two NLSs, and an Asp/Glu-rich acidic region (aa 434-572). One splice variant is known that shows a 15 aa substitution for aa 696-740. Human Daxx shares 71% and 90% aa identity with mouse and dog Daxx, respectively.
