Lipocalin-2/NGAL (Neutrophil Gelatinase-associated Lipocalin) is a secreted carrier protein that acts in the innate immune response, differentiation, tumorigenesis, and cell survival. It is induced in macrophages, glial cells, and epithelial cells during inflammation, anemia, and hypoxia. Lipocalin-2 binds catecholate siderophores which are secreted by bacteria and are required for bacterial iron uptake. Lipocalin-2 functions as a bacteriostatic agent in the innate immune response by limiting the bacterial iron supply. Iron uptake by Lipocalin-2 into mammalian cells is important for the regulation of iron-sensitive gene transcription. In the kidney, Lipocalin-2-mediated iron trafficking is required for protection from renal injury.

The Quantikine™ QuicKit™ Human Lipocalin-2/NGAL Immunoassay is a one step, 80-minute solid phase ELISA designed to measure human Lipocalin-2 levels in cell culture supernates, serum, heparin plasma, saliva, and urine. It contains NS0-expressed recombinant human Lipocalin-2 and antibodies raised against the recombinant protein. Results obtained using natural human Lipocalin-2 showed linear curves that were parallel to the standard curves obtained using the QuicKit standards. These results indicate that this kit can be used to determine relative mass values for natural human Lipocalin-2.

Serum/Plasma/Saliva/Urine - Samples from apparently healthy volunteers were evaluated for the presence of human Lipocalin-2 in this assay. No medical histories were available for the donors used in this study.
Cell Culture Supernates
Human peripheral blood mononuclear cells (1 x 106 cells/mL) were cultured in RPMI supplemented with 10% fetal bovine serum, 2 mM L-glutamine, 100 U/mL penicillin, and 100 μg/mL streptomycin sulfate. Cells were cultured unstimulated or stimulated with 10 μg/mL PHA. Aliquots of the cell culture supernates were removed and assayed for levels of human Lipocalin-2.
