Application Notes
WB: Approx. 50 kDa band observed in fetal mouse brain lysates and 35 kDa band plus fainter 50 kDa band bands in adult mouse brain lysates (calculated MW of 52.9 kDa band according to NP_061280.2). The observed 35 kDa band corresponds to the calculated MW of human isoform 2 (35.2 kDa band according to NP_001138597.1), but this size isoform has not yet been confirmed in mouse. In transfected HEK293 transiently expressing full-length human MKRN1 (myc and DYKDDDDK tagged), a band of approx. 60 kDa band was observed. No bands were observed in mock-transfected HEK293 and the same band was observed using anti-DYKDDDDK tag antibody.

Background: MKRN1

MKRN1 was identified as a repressor of c-Jun, androgen receptor, and retinoic acid receptor transcriptional activity. MKRN1 is a RING zinc finger protein that has been demonstrated to function as an E3 ubiquitin ligase. The repressive activity of MKRN1 is not dependent on its ubiquitin ligase activity. MKRN1 also appears to function as a transactivator of RNA polymerase II dependent transcription.
