Immunoprecipitation:MTHFD1 AntibodyNBP3-29959] -Detection of human MTHFD1 by western blot of immunoprecipitates. Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from 293T cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-MTHFD1 antibody NBP3-29959 (lot NBP3-29959-1) used for IP at 6 µg per reaction. MTHFD1 was also immunoprecipitated by rabbit anti-MTHFD1 antibodies for blotting immunoprecipitated MTHFD1, NBP3-29959 was used at 1 µg/ml. Detection: Chemiluminescence with an exposure time of 10 seconds.

Background: MTHFD1

MTHFD1 encodes a protein that possesses three distinct enzymatic activities, 5,10-methylenetetrahydrofolate dehydrogenase, 5,10-methenyltetrahydrofolate cyclohydrolase and 10-formyltetrahydrofolate synthetase. Each of these activities catalyzes one of three sequential reactions in the interconversion of 1-carbon derivatives of tetrahydrofolate, which are substrates for methionine, thymidylate, and de novo purine syntheses. The trifunctional enzymatic activities are conferred by two major domains, an aminoterminal portion containing the dehydrogenase and cyclohydrolase activities and a larger synthetase domain.
