Q: The molecular size of NEK8 is 75kDa. If you look at the WB image on the web for NBP1-82539(NEK8 antibody), the band is located between 70-100kDa (Closer to 100kDa). In our WB testing, we also observed the band near 100kDa. Do you think that it is the band of NEK8?
A: The predicted size of NEK8 protein is 75 kDa. However, the actual observed size of the protein is dependent on the sample type. If the protein has undergone post-translational modifications, then the size of the protein band could be higher or lower than the predicted one. Without a positive control such as a purified protein or overexpressed lysate, it is very hard to confirm 100% whether the band is real or not. We have not specifically tested MCF7 cell line, however we suspect that the band observed by you is the post-translationally modified NEK8 protein running at higher than predicted molecular weight.
