Immunoprecipitation:Neurofibromin 1 AntibodyNBP3-29269] -Detection of human NF1 by western blot of immunoprecipitates. Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from 293T cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-NF1 antibody NBP3-29269 (lot NBP3-29269-1) used for IP at 6 µg per reaction. NF1 was also immunoprecipitated by rabbit anti-NF1 antibodies. For blotting immunoprecipitated NF1, NBP3-29269 was used at 1 µg/ml. Detection: Chemiluminescence with an exposure time of 3 minutes.

Background: Neurofibromin 1

Neurofibromin 1 (NF1) is a suspected negative regulator of the ras signal transduction pathway. Neurofibromin is known to regulate the GTPase HRAS, causing the hydrolyzation and inactivation of GTP. Defects in NF1 lead to Neurofibromatosis disease, Recklinghausen disease or Watson disease. Neurofibromin has not been purified, therefore, most of the information regarding this protein has been deduced from homology analysis of its gene sequence.
