Immunoprecipitation:OAT AntibodyNBP3-29936] -Detection of human OAT by western blot of immunoprecipitates. Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from 293T cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-OAT antibody NBP3-29936 (lot NBP3-29936-1) used for IP at 6 µg per reaction. OAT was also immunoprecipitated by rabbit anti-OAT antibody BL21203. For blotting immunoprecipitated OAT, NBP3-29936 was used at 0.4 µg/ml. Detection: Chemiluminescence with an exposure time of 30 seconds.

Background: OAT

OAT, also known as mitochondrial enzyme ornithine aminotransferase, is a key enzyme in the pathway that converts arginine and ornithine into the major excitatory and inhibitory neurotransmitters glutamate and GABA. ( L-ornithine + a 2-oxo acid = L-glutamate 5-semialdehyde + an L-amino acid.) Mutations that result in a deficiency of this enzyme cause the autosomal recessive eye disease Gyrate Atrophy. Recombinant OAT protein was expressed in E.coli and purified by using conventional chromatography techniques.
