Specificity

The antibody detects a 75 kDa band on SDS-PAGE immunoblots of human PAK6 recombinant protein. This reactivity is specifically blocked by pre-incubation with PAK6 (N-terminal) peptide and is not affected by lambda phosphatase treatment.

Western Blot: PAK6 Antibody [NBP3-23282] - Western blots of human PAK6 recombinant protein phosphorylated by ERK2. The blot was exposed to lambda phosphatase (lanes 2 & 6) then probed with anti-PAK6 (N-terminal) (lanes 1-4) or anti-PAK6 (Ser-165) phospho-specific (lanes 5-8). The antibodies were used in the presence of unrelated (lane 3) and PAK6 (N-terminal) (lane 4) peptide or PAK6 (Ser-165) (lane 7) and unrelated phospho-serine (lane 8) peptides, respectively.

Background: PAK6

PAK6, a novel AR-interacting protein, shares a high degree of sequence similarity with other p21-activated kinases (PAK). PAK6 co-translocates into the nucleus with AR in response to androgen. In addition, AR and ER alpha transcriptional activites were inhibited by PAK6 in transient transfections with episomal and integrated reporter genes. PAK, which is highly expressed in testis and prostate tissues, could contribute to the effects of tamoxifen in breast cancer and in other tissues.
