Western Blot: Peripherin Antibody [NB300-137]

Western Blot: Peripherin Antibody [NB300-137]

Western Blot: Peripherin Antibody [NB300-137] - Analysis of tissue and cell lysates using Peripherin antibody, dilution 1:10000 in green: [1] protein standard, [2] rat spinal cord, [3] mouse spinal cord, [4] pig spinal cord, [5] cow spinal cord, [6] SH-SY5Y cells and [7] PC12 cells. The major band at ~57kDa corresponds to the major peripherin protein isoform, while other bands presumably represent protein products of alternate transcripts of the peripherin gene.

Immunocytochemistry/ Immunofluorescence: Peripherin Antibody [NB300-137]

Immunocytochemistry/Immunofluorescence: Peripherin Antibody [NB300-137] - Analysis of the widely used rat pheochromocytoma PC12 cell line stained with rabbit polyclonal Periphren antibody, dilution 1:2000 (Green). Hoechst staining of nuclear DNA (Blue). Peripherin, one of the Class III family of intermediate filament subunit proteins, is a major component of the PC12 cell forming a perinuclear cap, with some filaments visible in the cytoplasm.

Western Blot: Peripherin Antibody [NB300-137]

Western Blot: Peripherin Antibody [NB300-137] Detection of the 57 kDa peripherin protein in whole rat brain stem homogenate using NB 300-137 (1:20,000). The faint 48 kDa band is a peripherin molecule.
Immunocytochemistry/ Immunofluorescence: Peripherin Antibody [NB300-137]

Immunocytochemistry/ Immunofluorescence: Peripherin Antibody [NB300-137]

Immunocytochemistry/Immunofluorescence: Peripherin Antibody [NB300-137] - Mixed neuron/glia cultures from newborn rat brain stained with NB300-137 antibody to peripherin (green) and chicken polyclonal antibody to phosphorylated NF-H (red channel). A class of large neurons, like the one at the top right of this image, contain peripherin, while the majority of neurons and their processes contain NF-H and not peripherin. The blue channel reveals the localization of DNA.

Background: Peripherin

Peripherin (PRPH) is a 57kD type III neuronal intermediate filament (NIF) protein which is predominantly expressed in neurons of PNS and in certain CNS neurons specifically from brain stem/spinal cord which generally project toward peripheral structures. Inside developing neurons, PRPH is expressed abundantly in the growth cones and is required for axonal outgrowth as well as maintenance or regeneration. In mature neurons, together with other NIFs, PRPH constitutes a shape-maintaining IF network. Besides its crucial role in development of nervous system, PRPH can also be highly relevant to pathogenesis of neurodegenerative disorder. PRPH expression is induced in CNS neurons (wherein it is not expressed normally) upon brain injury, and factors such as LIF, IL-6 etc and PRPH molecules form aggregates in neurons. Synaptic plasticity is markedly altered in PRPH transgenic mice (Per mice) and up-regulation of PRPH expression is implicated in some pathophysiological conditions, such as cerebral ischemia and seizure, and some cognitive disorders. In ALS patients, PRPH together with neuronal IFs are detected in the majority of abnormal IF inclusion bodies in mature or aging motor neurons.
