The blue is DAPI staining of nuclear DNA. NBP1-05423 antibody reveals cytoplasmic filamentous staining in the small PC12 cells, while vimentin antibody stains intermediate filaments in the surrounding fibroblastic cells which do not express peripherin.

Immunocytochemistry/Immunofluorescence: Peripherin Antibody [NBP1-05423] - shows rat mixed neuron/glial cultures stained with NBP1-05423, our chicken polyclonal antibody to peripherin (green channel) and rabbit antibody to the neurofilament subunit alpha-internexin NBP1-05207 (green channel). These cultures contain mostly neurons which are rich in alpha-internexin, and a subgroup which have a large amount of peripherin also, such as the prominent cell in the middle of the micrograph. Since this cell expresses large amounts of peripherin and alpha-internexin, the green and red signals superimpose to produce a golden cell. Blue is a DNA stain.

Background: Peripherin

Peripherin (PRPH) is a 57kD type III neuronal intermediate filament (NIF) protein which is predominantly expressed in neurons of PNS and in certain CNS neurons specifically from brain stem/spinal cord which generally project toward peripheral structures. Inside developing neurons, PRPH is expressed abundantly in the growth cones and is required for axonal outgrowth as well as maintenance or regeneration. In mature neurons, together with other NIFs, PRPH constitutes a shape-maintaining IF network. Besides its crucial role in development of nervous system, PRPH can also be highly relevant to pathogenesis of neurodegenerative disorder. PRPH expression is induced in CNS neurons (wherein it is not expressed normally) upon brain injury, and factors such as LIF, IL-6 etc and PRPH molecules form aggregates in neurons. Synaptic plasticity is markedly altered in PRPH transgenic mice (Per mice) and up-regulation of PRPH expression is implicated in some pathophysiological conditions, such as cerebral ischemia and seizure, and some cognitive disorders. In ALS patients, PRPH together with neuronal IFs are detected in the majority of abnormal IF inclusion bodies in mature or aging motor neurons.

    Peripherin staining (green) in the mouse brainstem including the spinal trigeminal tract (right side of image) and arching fiber bundle to the nuc. solitary tract (center). Nissl Counterstain.
    Tested on both human and mouse tissues. Works well for staining of small diameter nerve fibers in peripheral tissues. Less effective in staining central terminals of sensory axons (mouse) but still effective (see image).
