Immunoprecipitation:PITRM1 AntibodyNBP3-29462] -Detection of human PITRM1 by western blot of immunoprecipitates. Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from 293T cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-PITRM1 antibody NBP3-29462 (lot NBP3-29462-1) used for IP at 6 µg per reaction. PITRM1 was also immunoprecipitated by rabbit anti-PITRM1 antibody. For blotting immunoprecipitated PITRM1, NBP3-29462 was used at 0.4 µg/ml. Detection: Chemiluminescence with an exposure time of 10 seconds.

Background: PITRM1

ATP-independent protease that degrades mitochondrial transit peptides after their cleavage. Also degrades other unstructured peptides. Specific for peptides in the range of 10 to 65 residues. Able to degrade amyloid beta A4 (APP) protein when it accumulates in mitochondrion, suggesting a link with Alzheimer disease. Shows a preference for cleavage after small polar residues and before basic residues, but without any positional preference
