Biological Activity

Protein synthesis inhibitor; leads to the premature release of polypeptide chains as polypeptidyl purine derivatives. Analog of the 3' end of aminoacyl-tRNA. Aminonucleoside antibiotic. Inhibits translation in both in vitro and in vivo systems. Also inhibits the transport of proteins into the mitochondria in vitro.

Puromycin maintains stable expression of recombinant hRyR2 in HEK293 cells.

Puromycin (5 mcg/ml) along with geneticin (250 mcg/ml) was used in DMEM containing 10% FBS, 4 mM glutamine and 10 mM HEPES to maintain the stable expression of hRyR2-IRES-GFP in HEK293 cell culture. The stable expression of hRyR2 in these cells was confirmed by cytosolic GFP fluorescence and 10 mM caffeine-evoked cytosolic Ca2+ peak, measured by Fura2 ratiometric dye, as shown in figure.
